Oral squamous cell carcinoma (OSCC) is the primary malignant lesion of the mouth, accounting
for over 95% of cases. Despite advances in surgical techniques as well as radiotherapy and
chemotherapy, the prognosis of the disease has not improved, with low survival rates. The most
used method in the prognostic evaluation of squamous cell carcinoma takes into account the
TNM system, where (T) is the lesion size, (N) is the infiltration of metastatic lymph nodes, and
(M) is distant metastases, an analysis defined in 2002 by the Union International for the Control
of Cancer. However, this method of evaluation has a limited predictive value. It is urgent
understanding of molecular biomarkers to clarify the clinical behaviour of this tumour in order to
individualise therapy in these patients. The aim of this study was to analyse
immunohistochemical staining of the protein NF-κB in OSCC tissue samples and evaluate the
potential correlation between expression status and clinicopathological features, as well as with
the patients survival or mortality. Our results showed no association between immunostaining of
NF-κB protein and overall survival of patients, data measured by the Log Rank Test (p = 0.727).
The chi-squared value proved to be significant in the histological grade (p = 0.036) and in
smokers (p = 0.037). These results confirm the involvement of NF-κB in oral carcinogenesis
associated tobacco smoke. Additionally, a significant increase in NF-κB levels correlated with
tumour dedifferentiation in OSCC, suggesting its association with disease progression.
